  • Embryo model

This collection highlights progress, challenges and discussions in the stem cell-based embryo model discipline. We welcome studies presenting new or refined models with improved cellular and morphogenetic complexity, studies utilising existing models to decipher mechanisms involved in early development, as well as relevant Reviews and Comments.

  • red and black cell

In this cross-journal Collection, we invite research article submissions focusing on non-immune and immune cell death, immune responses in health and disease, and the selective targeting of cell death regulatory molecules in the context of chronic inflammatory disorders.

  • Cells in green, red and blue

This cross-journal Collection showcases advances in understanding how senescence drives aging and age-associated disorders. We welcome studies investigating the cellular and regulatory mechanisms of senescence and its roles across tissues and disease.

    • Treatment with chemotherapy frequently results in the development of tumour resistance. A study now connects DNA damage in head and neck squamous cell carcinomas to cGAS–STING signalling, TNF production and paracrine-mediated stimulation of autophagy in neighbouring cells through enhanced activity of the ATG9A scramblase.

    • The human chorion provides the earliest structural foundation of the placenta, yet how it emerges from trophectoderm remains unclear. A recent study manipulates HIPPO–LATS–YAP signalling in transiently reset human embryonic stem cells to reconstruct a trajectory from trophectoderm to chorion in a dish.

    • Condensation of the helicase SMARCAL1 promotes chromatin accessibility, the resistance of tumours to radiation (through improved DNA repair), and T cell exhaustion (by altering methionine metabolism). Digoxin, a cardiac drug, represses SMARCAL1 condensates, restraining oesophageal cancer and improving responses to radiation and immunotherapy.

    • Dimethylation of the translation elongation factor eEF1A at lysine 55 by the dual methyltransferase METTL13 enhances decoding of a discrete set of low-frequency codons with weak wobble-position base pairing. This process selectively sustains synthesis of oncoproteins, including KRAS. METTL13 ablation restores sensitivity to inhibitors of the KRAS-G12C mutant and prolongs survival in models of lung adenocarcinoma.

    • Motile vesicles (MoVes) are a previously undescribed population of large extracellular vesicles released from the trailing edge of actively migrating cancer cells. MoVes carry the entire genome and mutational landscape of the parental tumour cell, together with intracellular organelles, including mitochondria, and proteins associated with epithelial-to-mesenchymal transition and organelle biology.

Stem cell-based embryo models

Because of their ability to simulate certain aspects of embryonic development, stem cell-based embryo models can be a promising tool in our efforts to better understand early embryogenesis, relevant disorders and fertility.
